Autophagy-targeted nanoparticles for effective cancer treatment: advances and outlook

چکیده
Abstract Autophagy enables the maintenance of nutrient recycling and metabolic homeostasis through a multistep lysosomal degradation pathway, it has been demonstrated that autophagy can act as tumor suppressor or promoter, depending on microenvironment (TME). The dual role in tumorigenesis results two opposing therapeutic strategies, namely, inhibition versus promotion. However, due to protective mechanisms cells absence specific strategies for regulation, modulation become major consideration cancer treatment. Owing their unique properties, nanoparticles (NPs) have excellent potential overcoming these limitations. Here, we provide summary latest progress autophagy-targeting NPs effective treatment, conclude with recent advances relevant clinical preclinical studies. This typical autophagy-targeted nano-drug delivery systems aims references expand ideas researchers intending explore this field. Finally, an outlook several key objective problems are carefully highlighted.

Targeted Nanoparticles for Cancer Therapy
The World Health Organization (WHO) estimates that 84 million people will die of cancer between 2005-2015 [1]. According using the WHO mortality database it has been estimated the total number of cancer deaths in the European Union (EU) and in 2012 is predicted to be 1283101, of which 717398 men and 565703 women [2]. The most common types of cancer that will be diagnosed are lung (C33-C34), int...

متن کاملSystemic Targeted Alpha Radiotherapy for Cancer
Background: The fundamental principles of internal targeted alpha therapy for cancer were established many decades ago.The high linear energy transfer (LET) of alpha radiation to the targeted cancer cellscauses double strand breaks in DNA. At the same time, the short range radiation spares adjacent normal tissues. This targeted approach complements conventional external beam radiotherapy and ch...
